Abstract

Dairy goat farming has great socioeconomic relevance in Brazil, especially in the Northeast region, and semen cryopreservation represents an essential tool for genetic improvement through artificial insemination. However, freezing and thawing processes induce oxidative stress, compromising sperm functionality. In this context, cannabidiol (CBD), a bioactive compound of Cannabis sativa with recognized antioxidant properties, has been proposed as a potential additive in cryopreservation media. The objective of this study was to evaluate the effects of adding different concentrations of CBD to the extender on post-thaw goat semen quality. Four viable ejaculates were used, which were pooled, diluted with CBD at concentrations of 0, 0.5, 2.5, and 5 µM, packaged into straws, and subjected to conventional slow freezing. After thawing, the samples were evaluated for motility, sperm movement patterns using a computer-assisted sperm analysis (CASA) system, and plasma and acrosomal membrane integrity. The results showed that the addition of CBD did not promote improvement in sperm quality, with a reduction in total and progressive motility, impairment of kinematic parameters, and a significant decrease in plasma and acrosomal membrane integrity in the treated groups compared to the control. It is concluded that, at the evaluated concentrations, CBD did not exert a protective effect during goat semen cryopreservation, highlighting the need for caution in its application and for further studies to elucidate safe concentrations and alternative strategies for its use.
